dc.descriptionABSTRACT This study is the first step in conducting molecular identification of sibling species of Anopheles punctulatus group of Papua. Research objectives were to develop molecular identification ofsibling species of An. punctulatus group of Papuans by using PCR-RFLP based on rDNA ITS2. The samples used were mosquitoes caught by the bait of people in East Koya and Arso II and An. farauti derived from a laboratory colony in BATAN Jakarta. Include the isolation of DNA molecular identification of mosquito and ITS2 rDNA amplification using primer ITS2A andITS2B by using PCR machine. ITS2 PCR results electrophoresed on 1.5% agarose gel containing 0.5 ug / ml ethidium bromide and observed using a UV transilluminator and photographed using a digital camera. Further amplification of ITS2 cut with MSP I restriction enzyme to produce a specific pattern of fragments that can be used for species identification. ITS2 amplification results for all samples of mosquitoes indicates the size of 750 bp. The results of identification by PCRRFLP for the mosquito An. farauti from laboratory colonies showed that laboratory colonies of mosquitoes were An. farauti ss formerly known as An. farauti 1. PCR-RFLP method developed in this study are expected to be cheaper and faster to use in species identification. Key words : Anopheles punctulatus group, molecular identification, ITS2 rDNA, sibling species , Papua. ABSTRAKPenelitian ini merupakan langkah awal dalam melakukan identifikasi molekular spesies sibling anggota Anopheles punctulatus group dari Papua. Tujuan penelitian untukmengembangkan identifikasi molekular spesies sibling anggota An. punctulatus group dari Papua dengan menggunakan PCR-RFLP berdasarkan ITS2 rDNA. Sampel yang digunakan adalah nyamuk yang ditangkap dengan umpan orang di Koya Timur dan Arso II serta An. farauti berasal dari koloni laboratorium di BATAN Jakarta.  Identifikasi molekular meliputi  isolasi DNA nyamuk dan amplifikasi ITS2 rDNA dengan menggunakan primer ITS2A dan ITS2B dengan menggunakan mesin PCR. Hasil PCR ITS2 dielektroforesis pada gel agarosa 1,5 % yang mengandung 0,5 ug/ml etidium bromida dan diamati dengan menggunakan UV transilluminator dan difoto dengan menggunakan kamera digital. Selanjutnya hasil amplifikasi ITS2 dipotong dengan enzim restriksi Msp I untuk menghasilkan pola fragmen spesifik yang dapat digunakanuntuk identifikasi spesies. Hasil amplifikasi ITS2 untuk semua sampel nyamuk  menunjukkan ukuran sebesar 750 bp. Hasil identifikasi dengan  PCR-RFLP untuk nyamuk An. farauti darikoloni laboratorium menunjukkan bahwa nyamuk koloni laboratorium tersebut adalah An. farauti s.s yang sebelumnya dikenal dengan nama An. farauti 1. Metode PCR-RFLP yang dikembangkan dalam penelitian ini diharapkan lebih murah dan cepat digunakan dalam identifikasi spesies.   Kata kunci : Anopheles punctulatus group, identifikasi molekular, ITS2 rDNA, spesies sibling, Papua.id-ID
